    Title: 探討不同光強度對Penicillium brevicompactum在液態發酵中生產Mycophenolic acid之影響

    Abstract: 植的歷史揭開序幕之後,越來越多的病人重獲新生,但器官排斥的問題一直是病人的一大考驗,許多接受移植的病人因自身的免疫能力造成器官急性排斥而必須把移植的器官取出,這直接威脅到病人的生命。因應而生的就是抗排斥的藥物,這也是醫界一直致力於解決的問題。而本論文就是利用Penicillium brevicompactum 來產生 Mycophenolic acid (MPA) ,一種用於器官移植抗排斥的藥物。 本篇論文的主要目的是在探討「光」這個環境因子對於P. brevicompactum生產MPA之影響。主要是探討的強度是從一般日常所見的日光燈因著照光的距離而調整其照度,主要的範圍是從0、400、1500、2500、3500 Lux;然而產物MPA的量則隨著照度的增加而隨著增加,達到最適化1500 Lux而後逐漸遞減。當操作條件為最適化時MPA的產量為照度0 Lux時的1.8倍(g/l),比生長速率也提昇約1.6倍之多,然而productivity則是提升了約2.6倍之多。隨後再增加照光強度則會逐漸遞減,但整體而言還是比照度=0 Lux時的好。 整體而言「光」對於P. brevicompactum 生產 Mycophenolic acid 是有正面幫助的,有照光的生長速率均比未照光的好,產物方面也是有一定的幫助。本實驗得到的結論為當照度為1500 lux 時所得到的結果是最佳的。無論是菌重、生長速率及產物的量都是達到一個最高值。 Effect of light intensity on the formation of Mycophenolic acid in the fungi Penicillium brevicompactum.In this study,we suggest that P. brevicompactum include many kinds of photoreceptor.To prove this suggestion,we used different light intensity ,0 Lux 400 Lux 1500 Lux 2500 Lux 3500 Lux, to discuss the growth of cell and the product .Over 200 hours,we can find the optimal environment for P. brevicompactum is at the light intensity 1500 Lux. In the optimal operation ,the biomass is 2g/l greater then at 0 Lux;the product is two times at 0 Lux. Another result is light will effect the category of growth. As we augment the light intensity, P. brevicompactum will change the category from mixd-growth associated product to no-growth-associated product.
