Specific aims: Development of screening tests for residues of beta-agonist in animal products. Important works in this year: (1)Collection of all informations about drug-residue detection techniques, products and key-raw components (monoclonal and polyclonal antibodies and hapten derivatives) to generate library. (2)Preparation of derivatives of beta-agonists. (3)Preparation of conjugate complexes of beta-agonist derivatives to different carrier proteins. (4)Immunization of mice for producing monoclonal antibodies. (5)Establishment of ELISA system to screen antiserum and monoclonal antibodies. (6)Cell fusion and screening of monoclonal antibodies. Select monoclonal antibodies against beta-agonists. (7)Purification and characterization of monoclonal antibodies.
Expected Results: (1)Establish the library related to the drug-residue detection techniques, products and key-raw components (monoclonal and polyclonal antibodies and hapten derivatives). (2)Produce beta-agonist hapten derivatives-conjugates 50 mg. (3)Produce beta-agonist protein-conjugate antigens 50 mg. (4)Animal immunization. (5)Establish specific ELISA system to screen monoclonal antibodies. (6)Produce monoclonal antibodies against beta-agonist. (7)Harvest and characterize pure monoclonal antibodies.
