    Abstract: 發展新一代高靈敏nSi 質譜分析晶片來提高藥品與治療成果間的對應性,利用各種表面處理之方式將nSi 質譜分析晶片上區域分別處理成hydrophobic、hydrophilic、cation exchange、anion exchange、metal affinity (IMAC)、DNA、antibody 與antibody 等不同表面特性的質譜分析晶片。如此 nSi 質譜分析晶片表面能同時於不同區段吸附捕捉不同特性之分子,將複雜之生物檢體分離於不同的質譜分析區段,進而增加質譜分析之選擇性 (selectivity) 以及可靠度 (confidence level)。固能將不同的生物分析過程如 on-chip digestion, desalting 或多維 (multi-dimensional) 分離模式如 size-exclusion chromatography, affinity chromatography, iso-electric focusing IEF 整合至單一晶片以多流道 (multiplexed) 平行進行分析以達到高處理量 (high throughput) 分析過程。本計劃並將整合國泰醫學中心獲臺灣及美國專利之人類胎盤幹細胞研究團隊,比較本群體計劃所發展的新一代高靈敏nSi 質譜分析晶片與傳統細胞學及蛋白質體學所測定、胎盤幹細胞在分化成不同神經細胞時的相關生物標記之一致性,以檢視此一高靈敏nSi 質譜分析晶片是否能有效應用於轉譯醫學的研究。 To develop the new nSi mass-analytical chip can promote the medical treatment and medical equipment industry. This study utilizes various methods to process the chip surface and froms different nSi mass-analytical chip, such as hydrophobic、hydrophilic、cation exchange、anion exchange、metal affinity (IMAC)、DNA and antibody chip. It can improve the selectivity and confidence level in mass analysis. This study also integrate the different bio- separating process, such as on-chip digestion, desalting or size-exclusion chromatography, affinity chromatography, iso-electric focusing, to achieve the high throughput. We will further apply the human placenta-derived multipotent cells, the stem cells identified by Cathay General Hospital, for the verification of the efficacy of our nSi mass-analytical chip. We will compare the biomarkers identified using tools of tranditional cell biology and proteiomics with those identified using our nSi mass-analytical chip during the induction of differentiation of human placenta-derived multipotent cells into different neuronal cells. This will provide useful information for the possible application of our nSi mass-analytical chip for future translational medicine. 研究期間:9903 ~ 9912
