    摘要: 環境檢測法中大腸桿菌的檢測需要較長時間培養、估算結果,若水體為生活休閒用水或飲用水水體,人體將曝露在高致病風險裡,因此檢測時間是一大問題,雖然已有不同快速檢測法開始著手解決,但都具有成本高、操作複雜等缺點,因此,本研究將利用吸收光譜法,使用酵素反應基質ONPG與大腸桿菌反應,藉由反應產物鄰-硝基酚具有吸光作用建立由分光光度計量測大腸桿菌之方法,藉以改善現在檢測法中耗時的缺點,而本研究建立之量測方法,僅需要加入反應基質0.5mM即可在35分鐘內得到尚可之線性相關量測模式,經由實際量測生活休閒用水體,35分鐘內量測到大腸桿菌數與環境檢測法中之酵素濾膜法比較差異百分比最低至最高為20.01%~53.40%,在量測數量方面,最低至最高可量測數範圍由49,300,000(CFU/100ml)~247,800,000(CFU/100ml),但在量測實際水體中,大腸桿菌可能因為有相同酵素之生物干擾而高估。
    The method of detection E. coli of environmental takes a long time to estimate the results, if water is a life of leisure or drinking, the human body will be exposed to a high risk of illness, and thus cause to detection time. Although there are different rapid detection method have issued, but all have high costs, complicated operation and other shortcomings, therefore, this study will use absorption spectrometry, using the E. coli enzyme reaction substrate ONPG reaction, the reaction product by o - nitrophenol with absorption spectrophotometer measured by the effect of establishing the method of E. coli in order to improve the current shortcomings of time-consuming assays, and the study established measurement methods, requiring only added to the substrate 0.5mM can get in 35 minutes the acceptable linear correlation measurement mode, by actual measurement life of leisure water, within 35 minutes, measured the number of E. coli and the environment in the enzyme assay of membrane filter percentage difference between the lowest to the highest was 20.01% ~ 53.40%, the measuring quantity, the minimum to the maximum number of measurements ranging from 49,300,000 (CFU/100ml) ~ 247,800,000 (CFU/100ml), but in the waste, sewage, E.coli may have the same enzymes as biological interference and overestimate.
