    摘要: 研究期間:10111~10210;Background; Elevating gas exchange efficiency in the culture medium is one of the goals for development of cell/tissue culture technology. System with high gas exchange can provide high amount of dissolved gas for the cells, and rapidly remove the exhaust generated from the cellular metabolism, yielding high cell proliferation. Furthermore, maximizing the use of supplied gas can significantly reduce the cost and therefore increase the value of products. However, it is difficult to have efficient gas exchange and high cell density simultaneously in a closed culture system by current technologies. To overcome this challenge, NCU/GIBME and Somapex Biotech Inc. has successfully exploited liquid fluorochemical as the gas carrier (2011-2012 / NSC 100-2622-E-008-013-CC3) in a photobioreactor and verified its effectiveness on microalgal growth. However, the availability of the previously developed system in practice may be hampered since (1) the system contained two immiscible phases, and (2) gas transport can only occur on the interface between fluoride and aqueous phases. After comprehensive review and discussion, the research team proposes to employ perfluorocarbon emulsions as gas carriers to establish a gas-exchange-enhanced photobioreactor for large-scale & enclosed cell culture use. Through the implementation of this project, we will not only extend the applications and values of fluoride materials, but further prompt biotechnological development in Taiwan and generate more benefits for the entire industry accordingly. Method and Features: In this study, the perfluorocarbon emulsions will be prepared using PEG-based fluorosurfactants produced by Somapex. The photobioreactor setup will be designed by NCU/GIBME. The effectiveness of the developed culture system will be verified by using Nannochloropsis oculata as the biological object. Several advantages can be obtained by using the developed system; 1. Emulsion particles can truly contact with cells that will dramatically increase the contact area between cells and gas carriers, yielding maximal efficiency of gaseous supply and transport within the system. 2. The developed bioreactor can be extensively used for animal and plant tissue/cell culture; especially for O2 and CO2-sensitive cell types, which is very helpful for biological / biomedical research. 3. Extend the application opportunities of fluorinated materials that will dramatically reduce the raw material prices, and whereby benefit all relative industries. Qualification/Capability; NCU/GIBME has robust research background in fields of bioengineering & cell biology, and has hands-on experience of fluorocarbon liquids handling in biotechnological applications. The partner; Somapex Biotech Inc. is a well-established biotech company with strong professions in fluorinated materials especially on fluorosurfactants (SBIR #1Z980534). This research team collaborated closely and presented outstanding research efforts in the past year, hence they are perfectly qualified to conduct this industry-academia collaboration program. Specific Aims : The aim of this one-year project is to establish a perfluorocarbon emulsions-mediated photobioreactor with high gas exchange efficiency for microalgal growth. The following tasks will be performed orderly; (1) Bioreactor designs, parameters configuration, and evaluation of different operation approaches. (2) Biocompatibility test of fluorosurfactants and perfluorocarbon emulsions generated. (3) Examination of efficacy of the developed photobioreactor on the cell culture. (4) Reproducibility examination and evaluation of the developed system for industrial use.
