    Title: 探討照光強度對納豆菌生長與納豆激酶生產之影響

    Abstract: 心腦血管疾病已經高居國人死亡原因的第二位,相關疾病包括血管硬化、心肌梗塞、腦中風、膽固醇過高、血脂肪過高等。而預防這些心腦血管慢性疾病,需要由飲食及生活型態來改變,其中配合膳食進行的改善,已成為世界流行的趨勢,而中國或西方傳統使用的草藥、機能性發酵食品或者具有歷史,以血管栓塞來說,日本傳統發酵食品-納豆便是最佳食品,在日本,「納豆」是1000年以上的養生食品,至今沒有發現副作用,故是一種安全的發酵食品,因納豆中含有血栓分解酵素—納豆激酶(Nattokinase)具有很強的血纖維蛋白〈fibrin〉溶解能力。 它主要是利用枯草桿菌(Bacillus subtilis)的類緣菌,俗稱「納豆菌」,以大豆原料進行發酵生成的一種蛋白質,它最重要的功能則是在於它是可以分解不溶性血纖維蛋白質的酵素,而近代生物科技已可由納豆中抽取納豆激酶,濃縮加工再強化其功效,可說是最有效預防血栓之方法。 對於納豆菌的相關報告來說,大多數是以固態發酵來進行,但是此一方法較不經濟,且易受雜菌污染,故擬以液態培養的方式來進行納豆菌的培養,並利用發酵槽進行擴大培養,以提高納豆激酶的產 量,探討光源強度從0 LUX到3500 LUX對於納豆菌以及納豆激酶產生的影響,由結果可以發現照度為0 LUX的時候,納豆激酶的活性只有4.22(FU/ml),其Y p/x為702.50(FU/g),其照度在1000 LUX有最大活性為18.29(FU/ml), Y p/x為3425.72(FU/g),所以可以證實Bacillus subtilis菌,雖然其屬於非光合菌,但是從參考文獻中證實菌中有光接收器進而會對於光產生反應,故對於產生納豆機酶有正面的影響。 To the relevant report in Nattokinase, the great majority go on by fermenting in solid state , but this method is relatively uneconomical, and apt to be polluted by the miscellaneous fungus , so plan to receive the cultivation of Bacillus substilis in way that the liquid fermentation , in order to add Nattokinase ,probe into light intensity of since 0 LUX to 3500 LUX for Bacillus substilis of and Nattokinase activity , when being can find that the intensity of light is 0 LUX by the result , Nattokinase activity 4.22 (FU/ml ),its Y p/x for 702.50 (FU/g),have intensity of light of it most activity not great for 18.29 at 1000 LUX (FU/ml), Y p/x is 3425.72 (FU/g ), so can verify Nattokinse is produced by Bacillus subtilis ,and the light intensity have a lot of influence. The suitable light intensity can produce the most Nattokinase activity.
