本研究以相位移相干涉術(Phase-Shifting Interferometry)結合表面電漿共振(Surface Plasmon Resonance)技術,由CCD擷取影像,配合區間積分資料擷取(Integrated-Bucket data acquisition)的方法,進行相位的檢測。由光路的設計與時域的相位重建分別達到系統的穩定性和動態能力。本系統由實驗可得相位解析度為0.4度,系統的折射率解析度為7.8 × 10^-6 RIU (Refraction Index Unit)。本系統具有即時、高靈敏、高解析度、免標定、大量平行篩檢等優點,預計可運用於微生物分子鍵結反應分析(biomolecular interaction analysis, BIA)的量測領域上,如抗原與抗體之交互作用、蛋白質分子的非特定吸附或DNA雜交等。A dynamic surface-plasmon-resonance imaging sensor was developed in this research. Phase-shifting interferometry with integrated-bucket data acquisition is employed to perform phase-image detection with a photodiode and a single CCD. System stability and dynamic capability were achieved through optical configuration design and temporal phase-extraction algorithm respectively. The resolution of phase detection was 0.4 degree, and the measurement sensitivity of refractive index was 7.8 × 10^-6 RIU (Refractive Index Unit). This system is capable of real-time measurements with high sensitivity, resolution, and throughput. It is anticipated to find applications in lable-free screening of biomolecular interactions, such as antibody-antigen interaction, nonspecific protein binding, and DNA hybridization.
