    摘要: 我們將沿襲以往的研究開創精神,開發合乎綠色化學的檢測技術,主要是利用快速與無需或微量有機溶劑(fast screening and solvent-free/less),市場上易購得的化學試劑及微萃取的概念,針對環境樣品、室內灰塵與生物檢體(如人類尿液、魚肉等),發展快速與少毒的檢測技術,建立一套方便、合乎環保又可靠的檢測技術,主要重點在樣品前處理上,不須購置特殊儀器設備、不須使用特殊化學試劑或吸附材料,並以前瞻性的微量檢測觀念,提出此三年型研究計畫。在頭兩年計畫中,將延續前次計畫中所提的新興污染物防腐劑- parabens (將增加檢測九種),再加上高產量化學品(HPV):Benzotriazoles, Benzothiazoles and Benzenesulfonamides 類為主要的待測物。以多變量實驗設計統計方式,於液態樣品前處理方面,評估不同微萃取方法的萃取效能,以微萃取技術(如Salting-out assisted liquid-liquid microextraction、Vortex-assisted liquid- liquid microextraction等),來探討液態樣品中含極性待測物萃取方法的最佳化條件。在固態前處理方面,以改良式基質固相分散萃取法(如Vortex- homogenized matrix solid-phase dispersion and Vortex-assisted matrix solid-phase dispersion),來探討固態樣品萃取方法的最佳化條件。在定性和定量上,開發氣、液相層析質譜技術(GC-MS and LC-MS)以檢測這類污染物質在不同環境與生物檢體中的殘留。於第三年的計畫中,我們根據頭兩年的研究結果,將探討以超效液相層析串聯高解析質譜(UHPLC-HR-MS)技術檢測這類污染物質在氯化消毒後的水環境中之代謝產物種類。此研究計畫將於檢測高產量化學品及新興污染物研究議題上,具有對檢測技術之突破性與研發上的主導性。我們亦將密切掌握國際上對於這類高產量化學品及新興污染物在環境與生物檢體中檢測技術的發展趨勢,以建立我國永續環境及食物鏈資料庫,並確保國人的健康及食品的安全。 ;In this three-year proposal, we will follow our last proposal to develop eco-friendly methods for the determination of four commonly used paraben preservatives plus five newly detected different sized ester side-chain and benzyl-parabens (total nine parabens) in various environmental, indoor dust and biota samples. Moreover, in this proposal, we will also develop eco-friendly methods to detect three groups of high production volume (HPV) chemicals: benzotriazoles, benzothiazoles and benzenesulfonamides, at the first time, in our environmental and biota matrices during the first two years. We will focus on developing solvent-free or solvent-less, and microextraction sample pretreatment techniques coupled with state-of-the-art GC-MS and LC-MS to analyze these target analytes. At the third year, based on the results from the first two years, the detection of chlorinated non-target metabolites of benzotriazoles and benzothiazoles in environmental water and tap water samples will also be developed and evaluated by the state-of-the-art UHPLC-HR- MS technique for the first time in this proposal. Followings four major tasks are going to be investigated and studied: Task 1. For liquid sample, evaluating salt-assisted liquid-liquid extraction (SALLE), vortex-assisted liquid-liquid microextraction (VALLME), Ultrasound- assisted emulsification microextraction (USAEME) and dispersive micro solid-phase extraction (DmSPE) for polar target analytes;Task 2. For solid, indoor dust and biota samples, evaluating modified vortex- homogenized matrix solid-phase dispersion (VHMSPD) and vortex- assisted matrix solid-phase dispersion (VAMSPE) and others; Task 3. Developing quantitative GC-MS, LC-MS with various UHPLC separation columns for target analytes;Task 4. Evaluating UHPLC-HR-MS technique to identify chlorinated non-target metabolites of benzotriazoles and benzothiazoles in environmental water and tap-water samples.The methods developed in this proposal significantly contribute to wide spectrum of “greener” sample pretreatment and microextraction techniques which also coupled with state-of-the-art hyphenated mass spectrometric for the determination of target and chlorinated non-target compounds in various environmental and biota samples. Eventually, we hope our newly developed methods can certainly be adapted with ease by routine analysis that may desire to track and examine residues of selected high production volume chemicals and emerging contaminants for human biomonitoring program. Moreover, the results of this research should be highly in response to our environmental protection trends, promote food safety and public health concern, as well as conservation and pollution control policies in Taiwan.
