    Abstract: 第二年目標:建立以生物活性成分為指標之發酵製程並試量產
    (a) 建立於巴西洋菇發酵製程中生物活性成份之分析方法
    例如建立potential biomaker “triterpenes”之分析方法
    (b) 針對不同生物活性作個別產程之最適化
    例如建立以Fed-batch發酵製程來增加potential biomarkers產量
    (c) 提供其他子計劃進行各種不同之生物活性分析與動物試驗
    與具有大型發酵槽之業者或研究機構合作生產提供其他子計畫進行各種不同之發酵樣品,以便利進行動物實驗(in vivo)與生物活性分析. 
    本計畫在執行期限內將可完成之工作項目如下:
    第二年
    (1) 建立巴西洋菇發酵製程中生物活性指標成份之分析方法。
    (2) 建立以生物活性成分為指標之Fed-batch發酵製程。
    (3) 建立以生物活性成分為指標之試量產發酵製程。
    (4) 提供其他子計劃進行各種動物試驗與生物活性分析。
    (5) 預期建立以生物活性成分為指標之發酵製程並試量產。
    Agaricus blazei have been confirmed as one of the most important medicinal and edible mushroom by many scientists. Previous research results indicate that the polysaccharides from the fermentation show the strongest anti-tumor activity among all known medicinal mushrooms in the animal experiments. Thus, it is important to develop the industrial fermentation processes for the anti-tumor polysaccharide production. Leguminous plants as fermentation substrates have been successfully exemplified by using soybean powder in fermentation industry in the past. However, there are some concerns regarding the functionality of the mycelium products from the fermentation. Besides, there are other leguminous plants have been found to have higher functional activities as compared that of soybeans. Thus, a well organized research groups with seven PIs from three known Universities (National Taiwan University, National Central University, National Yang-Ming University) have been formed to investigate the leguminous plants such as Astragalus membranaceus and black soybean as substrates. Three major medicinal compounds in leguminous plants are polysaccharides, saponins, and isoflavoinds. It is expected to have complete health food claims of the fermentation products from this research output, which include antioxidation, immuno-modulation, hormone replacement therapy, and hepapropective activities. Objectives:This sub-project not only provide the experimental materials to other sub-projects but also try to set up the fermentation strategy to enhance the functional metabolites from the fed-batch fermentation. Fed-batch technique has been widely used in the fermentation industry for the secondary metabolite production. However, there is little achievement in Agaricus blazei. In the preliminary study in our laboratory, we have found the operation conditions for the production of metabolite of Agaricus blazei. In search of the composition of the feed and the timing of the feed would be the major goals in this project.
    Project Structure :
    1. Building up the analytical methods of biomarkers to monitor the fermentation process of Agaricus blazei
    2. Optimizing the fementation process according to the biomarkers
    3. Pilot scale fermentation of Agaricus blazei 4. Supplying the high bioactive materials to other sub-projects for in vivo experiments
    Accomplishments:It is expected to accomplish some contributions toward the industry in this project.
    1. To obtain a high bioactive fermentation process of Agaricus blazei
    2. To obtain a pilot scale production process of Agaricus blazei with high bioactivity
    I believe that both the industry and academia will be benefited from the project, and the consumers will be the final winners.
