Abstract: This study explores the feasibility of using triblock copolymer polyampholytes as a displacer to separate beta-lactoglobulin A and B isoforms by anion exchange displacement chromatography. The displacers used herein are methacrylic polyampholytes: A(8)M(12)B(12), A(16)M(12)B(12) and A(16)M(24)B(24), where A is methacrylic acid, B is 2-(dimethylamino)ethyl methacrylate, and M is methyl methacrylate. In addition, the steric factor, characteristic charges and binding equilibrium constant with the ion exchanger gel of the triblock copolymer are measured and discussed with the structure and solution behavior of the polyampholytes. With the Steric-Mass Action (SMA) model of ion-exchange displacement chromatography, the elution and displacement line can be constructed for the system. Moreover, the behavior of elution and displacement line is qualitatively discussed and successfully proven.
