摘要: | 衣康酸由於其獨特的結構特性,因此廣泛地被利用在人造纖維、樹脂與其他合成材料上。而土麴黴 Aspergillus terreus 由於具有較高的衣康酸產率,因此成為許多研究機構討論的重點之一。由於過去文獻對於Aspergillusterreus 之研究主要是針對溫度、pH 值與通氣量等進行探討,對於光照的影響著墨不多,因此本研究遂以光照條件做為實驗之主軸。 本研究首先找出實驗所用菌株Aspergillus terreus BCRC 31128 之最適發酵溫度40℃,再設計以避光的0 Lux 為對照組,並探討在白光強度300、500、800 及1000 Lux,以及紅光與藍光150、300 及500 Lux 的條件下,對於衣康酸發酵的影響。 由實驗結果顯示,在白光強度500 Lux 以下的條件,對於衣康酸產量有正面幫助,但當白光強度高於800 Lux 時,則會對菌體的生長有抑制性。而低強度的紅光與藍光,雖然對菌重的生成沒有顯著影響,但皆比白光有較佳的刺激衣康酸生產效果;最佳之光照條件為強度300 Lux 的紅光,於此條件下,衣康酸產量比起避光條件要提升108 %,同時產物對基質的轉化率提升97.5 %。 本實驗發現單一波長之光源對於刺激Aspergillus terreus 生產衣康酸有較佳的效果,而紅光條件又優於藍光條件。 Itaconic acid, also called methylene succinic acid, is an important monomer for the manufacturing of resins, fibers, plastic, or other polymeric materials and has been widely utilized in chemical industries. Aspergillus terreus is one of the best itaconic acid producer because of its high yield. In past scientific literature, some experiment conditions, for instance, pH and temperature, are investigated. However, the investigation of light conditions for itaconic acid fermentation is deficient. For this reason, the study of mine discussed the effect on light quality for itaconic acid fermentation. In the study, the first step I found the best fermentation temperature is 40℃ by Aspergillus terreus BCRC 31128. Second, I devised the conditions, as the lack of light (0 Lux), and the conditions of 300, 500, 800, and 1000 Lux of white light. After all of the experiment of white light, I used red light and blue light, the conditions as 150, 300, and 500 Lux, and then discussed the results of all experiment. From the results, the yield of itaconic acid increased with the white light intensity below 500 Lux, and decreased with the white light intensity upon 800 Lux. The best light condition of itaconic acid fermentation is red light with intensity as 300 Lux, the amount of itaconic acid is 108% more than the condition of 0 Lux. |
